Composition and mechanism of action

The presentation of Orgalutran consists of a container that can be of two different formats:

  • Box with 1 disposable pre-filled syringe with a dose of 0.25 mg / 0.5 ml.
  • Box with 5 disposable pre-filled syringes of 0.25 mg / 0.5 ml dose.

Each syringe includes a needle protected with a natural rubber cap. The excipients of the aqueous solution are the following: acetic acid, mannitol and water for injectable preparations.

Orgalutran for ovarian stimulation

The active substance in Orgalutran is ganirelix. This is an antagonist of GnRH, which is a hormone synthesized in the hypothalamus and exerts its effect on the pituitary gland to induce the secretion of gonadotropins FSH and LH. This is achieved by activating the menstrual cycle of the woman and the ovarian follicles begin to develop to then expel a mature ovule.

In an assisted reproduction treatment, it is tried to avoid the occurrence of spontaneous ovulation and also to be able to control the ovarian stimulation thanks to the administration of drugs with exogenous hormones.

This is the function of ganirelix, inactivate the release of the endogenous hormones FSH and LH, in order to prevent premature ovulation of the follicles.



Indications and method of administration

Orgalutran is indicated in women undergoing assisted reproduction treatment, especially IVF, to avoid the endogenous peak of LH that occurs before ovulation and thus not lose the ovules before the follicular puncture.

This medication is used in combination with other drugs whose function is to stimulate multiple follicular development such as, for example, Menopur, Gonal-f or Puregon.

For the correct application of Orgalutran, the following instructions for use can be taken into account:

  • Injection subcutaneously, preferably punctured in the thigh.
  • The injection should be administered once a day at the same time.
  • Be sure to remove air from the syringe before injection.
  • Vary the injection site every day.
  • Do not give yourself an injection if the solution is not clear or contains particles.
  • Properly dispose of the syringe after use.

Treatment should begin between 96 and 120 hours after the start of ovarian stimulation, that is, on the fifth or sixth day of injecting the gonadotropins. This treatment should continue until the day of induction of ovulation, unless the doctor gives other indications.

The main difference that Orgalutran has with other GnRH analog drugs is precisely the time of administration. Being an antagonist, stimulation with Orgalutran consists of a short cycle.

Contraindications and precautions

The Orgalutran must not be used under any circumstances in the following cases:

  • Hypersensitivity to the active substance and excipients.
  • Allergy to the hormone GnRH or any of its analogs.
  • Women with moderate and severe alterations in renal and hepatic function.
  • Pregnant, lactating or postmenopausal women.

There are other cases in which you have to take special care before using Orgalutran as it could lead to serious effects on the person. Therefore, it will be necessary to consult with the doctor if it is possible to administer this medicine. These situations and warnings are the following:

  • Have an active allergy.
  • Latex allergy that contains the needle cap of this drug.
  • Risk of ovarian hyperstimulation syndrome.
  • Incidence of congenital malformations.
  • Risk of ectopic pregnancy with damaged fallopian tubes.
  • The efficacy of this drug in women weighing less than 50 kg or greater than 90 kg is not proven.

Orgalutran has to be in a refrigerator for its conservation?
